Ribonucleic acid synthesis at low and high temperatures in isolated winter wheat chromatin

Isolated winter wheat chromatin can synthesize heterogenous RNA with endogenous RNA polymerase. RNA synthesized with α-amanitin-sensitive RNA polymerase changed, quantitatively and qualitatively, during periods of germination at 24℃ and cold treatment at 4℃: (1) synthesis of medium (9 to 20S) mRNA at 24℃ decreased while that of small (5 to 9S) mRNA increased in the later stages of germination, and (2) synthesis of medium mRNA at 3℃ decreased while that of small mRNA increased during cold treatment. Chromatin from germinated shoots predominantly synthesized medium mRNA at 24℃ and small mRNA at 3℃. Chromatin from cold-treated shoots predominantly synthesized large (more than 20S) and medium mRNAs at 24℃ and small mRNA at 3℃. These results suggest that the ability to synthesize high molecular weight RNA at 24℃ is markedly stimulated by cold treatment of young shoots, and that some of the high molecular weight transcripts activated at 24℃ have an important role in vernalized wheat shoots transferred from 4℃ to 24℃ for further development.
