Basic and functional elements of Golgi apparatus

Mannosidase II (Man II) is one of Golgi-resident enzymes but shows rapid redistribution into endoplasmic reticulum (ER) under the effect of brefeldin A (BFA) followed by degeneration of the Golgi stack. This phenomenon is interpreted as the disappearance of the Golgi apparatus itself or diffusive mixing of Golgi and ER membranes. However, ultrastructural observation reveals that there remains membranous cluster, which is apparently distinctive from ER. We think this membranous cluster should be the basic or plastic element of the Golgi apparatus and Man II containing membrane is the functional or flexible one. GM130, a Golgi matrix protein, is detectable in not only intact Golgi apparatus but also BFA-induced Golgi clusters without dispersion into ER. After wash-out of BFA, reassembly of Man II on GM130-positive membranes and regeneration of Golgi stack can be observed. From these results, we concluded that most of membranous structure is the basic element of Golgi apparatus on which GM130 is closely associated and Man II is the functional and removable element. By addition of functional elements to basic ones, integrated structure and normal function of the Golgi apparatus could be established.
