Localization of Cathepsins B, D, L, LAMP-1 and μ-Calpain in Developing Hair Follicles

Hair follicles consist of multilayers encircling the core hair shaft, in which the hair cortex is surrounded by the hair cuticle. The encircling layers are broadly classified into internal and external root sheaths. The internal root sheath consists of cuticles of internal root sheath, Huxley's layer and Henle's layer. A layer lies between the external root sheath and Henle's layer is called the "companion layer". The hair bulb is the basal part of the hair follicle, and is composed of special tissues including dermal papilla, hair matrix, and precursor cells of the internal root sheath. Development of the hair follicle implies the coordinated differentiation of these tissues. During differentiation of tissues of the internal root sheath and hair shaft, most cellular organelles disappear and are replaced with keratin fibers. We conducted electron microscopic and immunohistochemical studies in hair follicles of the dorsal skin of neonate rats to investigate the roles of autophagy and proteolysis in this conversion. Autophagy was frequently found in the hair shaft during the keratinization process, especially in the hair cuticle area. Immunohistochemical study revealed the striking co-localization of cathepsin B, cathepsin D, cathepsin L, LAMP-1 and μ-calpain in the external root sheath, keratinizing Henle's layer, keratinizing hair shaft and primitive sebaceous gland suggesting the coordination of lysosomal proteases with cytoplasmic exzymes in promotion of the cytoplasmic conversion or terminal differentiation of hair follicle cells.
