The complex of chrome-azurol S (CAS) and human serum albumin (HSA) was dissolved by a nonionic detergent, Brij-35, and the solution was submitted to a spectrophotometry of HSA. The reagent solution was fixed as 2 : 1 : 1 mixture of 1.0 × 10^<-4> CAS, 0.3 % Brij-35, and 0.1 M lactic acid, and 4.0 ml of the reagent solution was added into 1 ml sample solution. After standing for 20 min, at a room temperature above 15℃, the absorbance at 550 nm was measured against the reagent blank. Beer's law was held from 50 ng to 250jig with the ε value being 3.1 × 10^5 1 mol^<-1> cm^<-1> closely to twice that of bromocresol green (BCG) method. While most of commom ions and anticoagulants gave no interferences, heparin, Al(III) and Fe(III) gave a large positive error. This method was applied to determination of HSA in 5 jj of normal control serum, control sera elevated lipid and bilirubin. The obtained HSA values were larger by about 15 % than those obtained by BCG method, indicating that globulins probably interfered the determination. However, the present results that GAS outside of sulfophthaleins interacts with HSA to extent of determining HSA in twice sensibility of BCG method gave a significant suggestion to develop a highly sensitive reagent for HSA.
