Stimulation of Microbial Conversion of Glycine into L-Serine by Magnesium Phosphate : Microbial Conversion of Glycine into L-Serin with Nocardia butanica(II)

Microbial conversion of glycine into L-serin was investigated with Nocardi butanica. The addition of tribasic magnesium phosphate [MG_3(PO_4)_3・8H_2O, t-Mgp], dibasic magnesium phosphate (MgHPO_4・3H_2O, d-MgP), and magnesium hydroxide in combinaiton with phosphoric acid to the fermentation medium was found to stimulate the L-serine produciton. The maximum level of L-serin was 10 mg/ml in 5 days (1.5 mg/ml of L-serine in the control experiment). Both glycine consumption and L-serine formation were initiated after glucose was almost completely consumed, which suggests taht glucose inhibits the utilization of glycine for L-serine formation.Concomitant with the L-serine formation, the pH-value of the culture medium rose, which was found to be due to the generation of ammonia. The increase of ammonium ion concentration and the rise of pH-value were suppressed by the addition of t-MgP, while the L-serine fromaiton continued. pH-Control of the culture medium by means of phosphoric acid did not enhance the L-serine formation as t-MgP did. The stimulation of L-serine produciton by t-MgP was also observed with N. paraffinica and Brevibacterium ketoglutamicum.
