Cultural Conditions for Microbial Conversion of Glycine into L-Serine in the Presence of Tribasic Magnesium Phosphate : Microbial Conversion of Glycine into L-Serine with Nocardia butanica (III)

Stimulation of microbial conversion of glycine into L-serine with Nocardia butanica by magnesium phosphated has been reported from this laboratory. Detailed cultural conditions wer studied for efficient L-serine production in the presence of tribasic magnesium phosphate (t-MgP). Less then 8% of carbon sources and low concentrations (less than 1%) of nitrogen sources were suitable. Addition of L-alanine, norleucine, or some organic solvents (acetone, ligroin) stimulated L-serine production. Growth inhibition observed at a high glycine concentration was reduced by adding glycine to the fermentation medium at the middle to late logarithmic phase of growth, resulting in a high yield of L-serine. Under optimal cultural condition, more than 12 mg/ml of L-serine was accumulated with a trace of residual glycine and without concomitant production of other amino acids. The molar conversion ration of glycine into L-serine was nearly 50% under the conditions employed.
