Recent studies have demonstrated that radiation damage in DNA is induced not only by direct and indirect effects but also by the quasi-direct effect. The quasi-direct effect of ionizing radiation on DNA is usually investigated in its frozen aqueous state at 77 K. In a previous study we demonstrated the usefulness of the spin-trapping method with α-phenyl-N-tert-butylnitrone (PBN) to study the quasi-direct effect and proved that OH radicals and hydrated electrons were generated in the hydration layer of DNA when exposed to γ-rays at 77 K. As the mechanisms of the quasi-direct effect, we proposed that hydroxyl (OH) radicals generated in the hydration layer of DNA participated in the induction of base alterations such as 7,8-dihydro-8-oxo-2'-deoxyguanosine (8-oxodG), but not in the formation of strand breaks. In the present study PBN was again employed to elucidate the roles of hydrated electrons in the formation of DNA damage by the quasi-direct effect of ionizing radiation.
