Infrequent Mutation of Waf1/p21 Gene, a CDK Inhibitor Gene, in Brain Tumors

PCR-SSPC法およびDNA sequence解析を用いて，脳腫瘍28例(astrocytic tumors15例，medulloblastoma6例，pineal parenchymal tumors5例，neuroblastoma2例)におけるWaf1/p21遺伝子の変異の解析を行った．Low grade astrocytomaの1例の一部のPCR cloneでcodon33のmissense mutationを，medulloblastomaの1例でcodon35のsilent mutationを認めたにすぎなかった．今回検索した脳腫瘍におけるWaf1/p21遺伝子の変異は稀であると考えられた．さらに今回の検索で，正常対照および脳腫瘍患者標本においてcodon31にAGC(Ser)/AGA(Arg)のpolymorphismを認めた．AGAを有するalleleの頻度は欧米のデータに比べてかなり高く，人種的背景によるものと考えられた．
