[SHORT REPORTS]DNA Binding Specificity of Thyroid Hormone Receptor Isoforms:Rol of the 9th Heptad Repeat

Thyroid hormone receptors (TR_s) are one of the members of steroid hormone receptor superfamily and are encoded by two different genes, α and β. In the TRα gene, three isoforms (α1, α2 and α3) can be created by alternative splicings that change the amino acid sequences at the critical dimerization region (9th heptad repeat). To evaluate the involvement of the 9th heptad in DNA binding and dimerization, gel mobility shift assays were performed. TEα1 formed a monomer, homodimer and heterodimer with the retinoid X receptor α on both the Lap and DR4 types of thyroid hormone-responsive elements. TRα2 and TRα3 bound to DR4 only as a heterodimer, whereas neither isoform bound to Lap. There results indicate that alterations in the 9th heptad repeat in TRα isoforms could affect the DNA binding specificities and might mosulate their functions in vivo.
