EFFECTS OF SEX STEROID HORMONES ON RAT ANTERIOR PITUITARY LH-RH RECEPTOR

ゴナドトロピン分泌調節における性ステロイドホルモンの下垂体レベルでの作用機序を明らかにする目的で,estradiolの下垂体前葉LH-RH receptor および下垂体前葉のLH-RH反応性に及ぼす影響を検討した.Wistar系の去勢雌ラットに1日20μgの17β-estradiol (E),2 gmのprogesterone (P),またはE+Pを投与し,摘出された下垂体をTris-MgCl_2-KCl bufferでhomogenizeし,比活性30Ci/mmolの^3H-LH-RHとncubate後,Millipore filterにて分離し,receptorを分析した.一方,下垂体前葉のLH-RH反応性は,同様に処理したラットに0.4μgの合成LF-RHを投与し,LHの反応をRIAにて測定して検討した.去勢雌ラット下垂体前葉のLH-RH receptorの解離恒数は2.08×10^<-8>M,結合部位数は27.6f mol/mg proteinであった.Eの2,4日間投与により,receptor 結合部位数は増加したが,6-10日間投与,またE+Pの10日間投与では減少し,P単独投与では変化がみられなかった.下垂体のLH-RH反応性はE投与により増加した.このように,estradiol 1日20μgの2,4日間投与では下垂体前葉のLH-RHに対する反応性とLH-RH receptor両者の変化が相関しており,エストロゲンは,下垂体前葉のLH-RH receptorの変化を介して下垂体前葉LH-RH反応性を調節していることが示唆された.
