Soybean Preference for Bradyrhizobium japonicum for Nodulation : Isolation of Rj_2Rj_4-Lines from the Cross of Soybean cvs. IAC-2 (Rj_2) and Hill (Rj_4)

To obtain soybean (Glycine max. L. Merr. ) lines harboring the Rj_2- and Rj_4-genes, the Rj_2-cultivar IAC-2 (Genotype Rj_2Rj_2rj_4rj_4) and Rj_4-cultivar Hill (rj_2rj_2Rj_4Rj_4) were crossed. Genotype Rj_2Rj_2Rj_4Rj_4 was selected based on its nodulation reactions with the combined inoculum of Bradyrhizobium japonicum strains Is-1 (nodulation-type B; nodulates the Rj_4-cultivar but does not nodulate the Rj_2-cultivar) and Is-34 (type C; nodulates the Rj_2Rj_3-cultivar but does not nodulate Rj_4-one). In the F_1 and F_2 generations, both Rj-genes segregated according to Mendel's laws. The genotype of the F_2 plant could be considered to be Rj_2Rj_2Rj_4Rj_4, if all the tested progenies from the F_2 plant nodulated ineffectively with the combined inoculum. Ten progenies from each of eight F_2 plants were assayed for the Rj-phenotypes and four promising F_2-plants were isolated. The progenies (F_3 generation) from the selected lines were transplanted in the field, and the F_4 progenies were examined for their compatibility with the combined inoculum strains. Three promising lines homozygous for both Rj_2- and Rj_4-genes and one line homozygous for Rj_2 and heterozygous for Rj_4 were isolated. The progenies (F_5 generation ) from the selected lines were examined for their compatibility with the combined inoculum strains, and it was eventually concluded that 12 lines (F_4 generation) were characterized by the genotype Rj_2Rj_2Rj_4Rj_4.
