Effects of Dexamethasone and Basic Fibroblast Growth Factor on the Differentiation of Rat Bone Marrow Stromal Cells

The purpose of this study was to determtine the capacity of dexamethasone(DEX)and basic fibroblast growth factor(bFGF)to promote the differentiation of rat bone marrow stromal(RBMS)cells in vitro. RBMS cells obtained from the bone marrow of 40-45 day-old Sprague-Dawley rats were grown in primary culture and then subcultured for up to 7 passages. After both DEX(10^<-8> M)and bFGF(3 ng/ml)were added to the cultures, the RBMS cells produced significant numbers of mineralized nodules. Similar resuits occurred with addition of bFGF alone. RBMS cells cultured both DEX and bFGF were highly stimulared in terms of cell proliferation, alkaline phosphatase activity and [^<14>C]proline incorporation, in comparison with the controis; surprisingly, bFGF alone was even more effective in producing these phenotypic changes. According to sodium dodecyl sulfatepolyacrylamide gel electrophoresis(SDS-PAGE)analysis, the cells secreted significant amounts of 70 kDa proteins in the presence of bFGF with or without DEX, compared with cell in the control cultures. There were no significant changes in the other protein profiles with or without these stimulants. These recults indicate thet bFGF plays a crucial role in the differentiation of RBMS cells, even though DEX has the capacity to stimulate expression of osteogenic parameters. This study may be the first to observe that bFGF has the capacity to stimulate calcification of nodules without DEX treatment.
