Cellular Expression of Alveolar Bone-Derived Cells Following Mineralized Nodule Formation

The present stydy describes alterations in the cellular expression of rabbit alveolar bone-derived cells (RAB cells) during in vitro mineralized nodule formation. We monitored alkaline phosphatase (ALPase) activity, secretion rates of collagenous proteins and collagen types at various intervals up to 21 days during culture, and examined the RAB cell phenotypes profile daily. The RAB cells, grown from bone fragments subjected to sequential bacterial collagenase treatment, were harvested and subcultured up to the 3rd passage. Nodule formation was initiated on day 7 of culture, when the cells became confluent. Mineralization of the nodules took place on day 10. The ALPase activity of the cells increased gradually up to day 10, then decreased markedly on day 12, coinciding with the initiation of mineralization. Protein synthesis was examined by [^<14>C] proline labeling and showed that [^<14>C] proline incorporation into the secreted proteins was higher on days 10 to 12 than on other days. Protein synthesisi on day 14 decreased to one third of that on day 12. The production rate of collagenous proteins compared to total protein synthesis gradually increased up to day 12. Extracellular processing of the α-chains of type I procollagen to collagen seemed to be accelerated, in parallel with the initiation of mineralization. Type III collagen production gradually decreased after the nodules mineralized. These results suggest that osteoblastic cells markedly alter their cellular expression during mineralization.
