β,β'-IMINODIPROPINITRILE INDUCED LIPID PEROXID ATION AND OXIDATIVE MODIFICATION OF PROTEIN

β, β'-Iminodipropionitril (IDPN) Produces a neurotoxicity recognized as the waltzing or the ECC (excitement, circling and choreic head motion) syndrome and a motor neuron degeneraton resembling, in part, findings in the central nervous system in cases of human motor neuron disease. As the neurochemical mechanism has not been fully clarified, we attempted to elucidate a process of peroxidation in the brain of IDPN-treated mice. IDPN (1g/kg b.w.) was administered by intraperitoneal injection every 3 days for a totl of 5 times. Results showed an induction of lipid and protein peroxidation. and a decrease in activity of superoxide dismutase, the scavenger enzyme for the superoxide anion as reactive oxygen activity, in the brain homogenate of IDPN-treated mice. This indicates that the motor neuron degeneration induced by IDPN may be due to oxidative stress of reactive oxygen, suggesting a possible contribution to the degenerative process in the central nervous system in human motor neuron disease.
