The loss of replicative potential observed in normal human cells in culture is well documented and utilized as a model for aging at the cellular level. Numerous changes in gene expression are known to occur during replicative senescense and multiple levels of regulation contribute to the irreversible growth arrest. One of the underlying molecular mechanisms in senescence is the impaired activation of both Cdk4 and Cdk2 whose activities are essential for the progression of the G1 phase. Recent progression in the study of mammalian cell cycle regulation may allow to explain the molecular events in senescent phenotypes.
