To determine the reaction of Cr salt with amino-acid radical quantitatively, the authors examined the optical characteristics of various solutions containing Cr and DNP-glycine. They also studied the behavior of combined Cr and combined DNP-glycine to the extraction with ether by means of a spectrophotometer. They devised a method to determine the amount of reaction between Cr salt and amino-acid radical, using DNP-glycine. The results obtained are summarized as follows.1. In a system consisting of SO2-reduced liquor and DNP-glycine solution, the Cr combined with DNP-glycine did not move from the Cr liquor to ether layer. The DNP-glycine combined with Cr, however, was not extracted with ether and remained in the Cr liquor. 2. The amount of DNP-glycine remaining in the Cr liquor after extraction increased proportionally, as the reaction proceeded between Cr and DNP-glycine. Hence the amount of reaction between Cr and amino-acid radical could be determined by the amount of the remaining DNP-glycine.3. The determination of the remaining DNP-glycine should be made by the following procedure: After lowering the pH of the mixture of Cr and DNP-glycine 2.5 below, the mixture was transfered to a separatory funnel. Subsequently it was shaken by hand with the same amount of ether for a few minutes. This extraction was performed twice at least. The absorbancy of the Cr liquor layer was measured by the spectrophotometer at a wavelength of 350mμ. The amount of the remaining DNP-glycine was calculated from the difference in absorbancies which had been measured before and after extraction.4. As the standard deviation found in the measurement of absorbancy was about 0.002, the amount of reaction, particularly at the early stage of reaction, between Cr and amino-acid radical was determined, with ease and reliability, by the method devised by the authors. This method, however, was unfavorable in the point that the ratio of Cr/DNP-glycine determinable by the method was limited.
