細胞膜糖脂質のナノスケール分布解析

生体膜は二次元方向の流動性を持つが、膜タンパク質、膜脂質ともに分布は一様ではなく、様々な集中や偏りを示すことが知られている。しかし膜タンパク質に比較して、膜脂質の分布を可視化する方法は限られ、内在性の分子を定量的に解析することは困難であった。我々は急速凍結と凍結割断レプリカによる物理的固定を用いて,ナノスケールで膜脂質の二次元的な分布を捉えることに成功した。この方法による膜脂質の捕捉効率は高く、空間統計学的な解析が可能である。この稿ではおもに細胞膜外葉のGM1, GM3に関する結果を紹介し、内葉のPI(4,5)P2についても触れる。
