HPLC Profile Analysis of Hepatoprotective Oleanene-Glucuronides in Several Soybean Processed Foods

HPLC analysis of oleanene-glucuronide (OG) was performed on the following commercially available soybean processed foods: Tohnyu (soymilk), Tofu (soybean-curd), Okara (soybean-curd lees), Age (fried-tofu), Koya-tofu (freeze-dried tofu), Kinako (soybean flour), Moyashi (soybean sprouts), Natto (fermented soybeans), Miso (bean paste), and Shoyu (soysauce). All the foods listed above have OG except for Miso and Shoyu, though in varying amounts. Furthermore, the HPLC profiles of the total OG fraction obtained from the foods were compared. There was no great difference in HPLC profiles with respect to different methods of processing. The structures of the major OGs were identified as soyasaponins I (1) and II (2). Total OG obtained from all these foods showed hepatoprotective effects towards immunologically induced liver injury in primary cultured rat hepatocytes. Total OGs in Tohnyu, Okara and Koya-tofu showed stronger hepatoprotection than the other foods, since these foods contained larger amounts of soyasapogenol A-type glucuronides than those in the other foods.
