Quantitative Determination of Aconitic Acid in Processed Aconiti Tuber

A simple and precise ion-control high-performance liquid chromatographic method was developed for the determination of trans- and cis- aconitic acid in processed aconiti tuber (TJ-3022). For this purpose, a reversed-phase chromatographic system consisting of a chemically bonded ODS silica gel column was used with a phosphate buffer (pH3.0) containing 5mM NH_4H_2PO_4, as a mobile phase. The isomers were separated and their contents were individually determined within 20min after injection of the test solution in 0.1%NaHCO_3/methanol(1:1). The analysis showed that the trans- and cis-aconitic acid contents in TJ-3022 were 0.72% and 0.25%, respectively.
