Analysis of Gastric Mucosus membrane-protective compounds in Coptidis rhizoma

• An HPLC method was established for analysis of coptisine and 8-oxocoptisine, (+)-lariciresinol and p-hydroxyphenethyl trans-ferulate of Coptidis rhizoma, which shows an activity of gastric mucosus membrane-protection. Their contents in several samples of Coptidis rhizoma were measured by the HPLC system, using an ODS reversed-phase column and a UV absorption-monitoring system at 345 nm and 280 nm. The crude drugs were shown to contain 1.1-1.7 % of coptisine, 0.006-0.009 % of 8-oxocoptisine, 0.003-0.005 % of (+)-lariciresinol, and 0.001-0.002 % of p-hydroxyphenethyl trans-ferulate. We examined the contributory ratio of coptisine, 8-oxocoptisine, and (+)-lariciresinol in the gastric mucosal protection and revealed that coptisine is mainly responsible for the activity of Coptidis rhizoma.
