野生型(AhR+/+)ならびにアリールハイドロカーボン受容体欠損(AhR-/-)マウス肝臓における2,3,7,8-Tetrachlorodibenzo-p-Dioxinによる遺伝子誘導プロファイル(毒性学)

• 2,3,7,8-Tetrachlorodibenzo-p-dioxin(TCDD)は,哺乳類に様々な生物学的影響を起こす最も毒性の強い環境汚染物質の1つである.本研究の目的は,TCDDにより誘発される肝毒性と肝癌に関係する遺伝子の同定である.TCDDの投与量は100μg/kgとし,C57BL/6(AhR+/+, wild type)およびB6.129-AhR<tmlBra>/J(AhR-/-, knock out)マウスの腹腔内に1回投与した.AhR+/+マウスにおいて,TCDD投与後72時間後で,明らかな組織化学的変化は認められなかったが相対肝重量は有意に増加した(P<0.05).また,AhR-/-マウスにおいて,TCDD投与により,血清alanine aminotransferase活性が有意に増加した(P<0.05).肝臓における遺伝子発現プロファイル解析を投与後72時間で行ったところ,AhR-/-マウスに比較して,AhR+/+マウスでは51遺伝子の発現に変化(>3倍)が見られた;28遺伝子は誘導され,23遺伝子は抑制された.これらの遺伝子のほとんどは,化学走性,炎症,発癌,急性相反応,免疫反応,細胞代謝,細胞増殖,情報伝達や癌抑制に関連性があった.本研究から,AhR+/+とAhR-/-マウス肝における遺伝子microarry解析は,TCDDによるAhR介在肝毒性ならびに肝癌の発現メカニズム解明に有用であることが示唆された.
