フローサイトメータによるエチレングリコールモノエチルエーテルのラットの精子形成に対する影響評価

フローサイトメータを用いて,ラットの睾丸細胞に対するエチレングリコールモノエチルエーテル(EGEE)の影響を調べた。EGEEをラットに0,100,200,400,800mg/kg/dayの投与量で4日間経口投与した。EGEE投与により睾丸重量,精巣上体,睾丸細胞数は減少した。組織学的観察には200mg/kg投与群において,性細胞の管内への剥離が見られた。400mg/kg投与群では睾丸の中等度の変性がおこった。フローサイトメトリーから,ハプロイド細胞の有為な減少と,ディブロイド細胞とテトラプロイド細胞の比率の異常が認められた。これらから,EGEEによる雄性生殖器に対する毒性は,睾丸の性細胞の比率の異常をもたらすと考えられた。
