Experimental Leptospirosis with Special Reference to Cell-Mediated Immunity

In leptospirosis in man, killed vaccine is effective against severe leptospirosis; and in rodents, killed vaccine and lipopolysaccharide specific to serotype are protective antigen in leptospiral infection. Prior to the current study, it was well known that leptospirae are detectable in the kidney at the convalescent stage of this immunity. In the present study, using macrophage migration inhibition test (MIT) and skin reaction, we demonstrated delayed type hypersensitivity in leptospiral infection of mice and guinea pigs. Guinea pigs immuized with Leptospira serovar icterohaemorrhagiae showed positive reaction to L. serovar ieterohaemorrhagiae and L. serovar canicola in MIT. Water extracts of infected liver cells showed positive reaction to the lymphocytes sensitized with L. serovar icterohaemorrhagiae, L. serovar autumnalis and L. serovar canicola. Skin reaction was also positive in guinea pigs immunized with L. serovar icterohaemorrhagiae. Similar results were obtained with mice.
