Effects of occlusal hypofunction on the microvasculature and endothelin expression in the periodontal ligaments of rat molars

The purpose of this study was to examine the effects of occlusal hypofunction on the vasculature and endothelin expression in periodontal ligament. Infusion of ink to visualize vascular changes and immunohistochemical processing for endothelin were used. 1. At 1 week post-resection, few large vessels and many small vessels were observed. Blood flow extended irregularly from the bone into periodontal ligaments, while such extention was regular in normal occlusion. At 3 weeks, the microvasculature in periodontal ligaments was even more irregular. 2. In the normal occlusion group, endothelin-like immunoreactivity was widespread in vascular endothelial cells and fibroblasts of periodontal ligaments. In the hypofunctional occlusion group, immunoreaction of endothelin in the periodontal ligaments was significantly stronger than the normal occlusion group at 2 days, 1 week and 3 weeks. Furthermore, the hypofunctional occlusion group showed a much stronger reaction in some small arterioles. These results suggest that endothelin may affect regulation of the periodontal ligament microvasculature under occlusal hypofunction.
