Effects of conditioned medium of continuously compressed Human Periodontal Ligament Fibroblasts on MC3T3-E1 cells

First, we attempted to investigate some effects of the conditioned medium of continuously compressed human periodontal fibroblasts (HPLF) on clonal mouse osteoblasts-like cells (MC3T3-E1 cells). After the application of compressive stress to HPLF, the conditioned medium of HPLF (S-CM) was collected. The conditioned medium of non-compressed HPLF (NS-CM) and the control medium without cell culture (C-M) were also collected. Both ALPase activity and cAMP production in MC3T3-E1 cells were assayed after an addition of S-CM, NS-CM or C-M. Furthermore, ALPase activity and cAMP production were measured when MC3T3-E1 cells were directly compressed, and the results were compared with those in the MC3T3-E1 cells in the presence of S-CM. Secondly, in order to determine whether prostaglandins (PGs) were associated with the effect of S-CM on MC3T3-E1 cells, S-CM including indomethacin was added to the medium containing MC3T3-E1 cells, and then ALPase activity and cAMP production were assayed. The results were as follows : 1. An addition of S-CM increased both ALPase activity and cAMP production in MC3T3-E1 cells time-dependently, as well as observed in directly compressed MC3T3-E1 cells. In the presence of S-CM as compared to NS-CM or C-M, significant increase in ALPase activity and cAMP production in MC3T3-E1 cells was recognized at 12 and 6 hours after treatment respectively. 2. In continuously compressed MC3T3-E1 cells, ALPase activity and cAMP production increased at 6 and 3 hours after treatment respectively. 3. S-CM in the presence of indomethacin increased neither ALPase activity nor cAMP production in MC3T3-E1 cells. These findings demonstrated that S-CM might have the ability to enhance the cellular activities of MC3T3-E1 cells, and that PGs seemed to have an important role in its phenomena.
