Osteopontin localization and expression in cellular cementum at the site of root resorption during physiological tooth movement

The objective of this study was to investigate the role of osteopontin (OPN) secreted by cementocytes in root resorption. The localization of OPN immunoreaction and mRNA in cellular cementum of rat molars during physiological distal drift were investigated. Furthermore, tartaric acid-resistant acid phosphatase activity was detected as a marker of odontoclasts and its precursor cells. At the distal side of the buccal roots of maxillary molars, many of cementocytes in cellular cementum facing actively resorbed alveolar bone were positive for OPN immunoreaction and OPN mRNA. Immunoreaction for OPN was also demonstrated at the cementocyte lacunae and canaliculi. Odontoclast precursors appeared on the cementum surface where OPN-positive cementocytes were localized. Odontoclasts were found in the vicinity of OPN-positive cementocytes. At the mesial side where no resorption was observed, OPN immunoreaction and mRNA were observed in few of cementocytes. Ultrastructural findings revealed that cell processes of cementocytes were in close contact with odontoclasts which were resorbing cementum. These results suggest that OPN secreted by cementocytes plays an important role in the induction of odontoclasts to the cementum surface.
