Experimental tooth movement alters the expression of TrkB-like immunoreactivity in the periodontal fibroblasts of rat molars

Our previous immunocytochemical study reported the expression of TrkB, a neurotrophin receptor, in various non-neural cells in rat periodontal ligament, though its functional significance remains obscure. In the present study, spatiotemporal changes in the distribution and immunointensity of TrkB-like immunoreactivity (-Ll) were examined in rat periodontal fibroblasts during experimental tooth movement by immunohistochemistry combined with computerassisted image analysis. Many periodontal fibroblasts exhibited TrkB-Ll both in the experimental and control groups. An image analysis demonstrated chronological changes in TrkB immunopositive pixels, reminiscent of TrkB immunoreaction products, in each experimental period. However, the ratio of the immunostained area to the periodontal ligament did not show significant differences throughout the experimental periods. These numerical data indicate that orthodontic forces altered the distribution of the TrkB-positive area, but not their number. On the other hand, the immunointensity of TrkB reached its maximum on Day 1 of treatment (p<0.05), and thereafter decreased gradually to recover to control levels on Day 7 of treatment. Since the involvement of neurotrophins has been reported in proliferation and differentiation on fibroblasts, TrkB might play a role in the tissue remodeling of the periodontal ligament incident to experimental tooth movement.
