Prevention of Fibrosarcoma HT 1080 Cell Detachment by Multimeric Fibronectin Formed in vitro

Cultured human fibrosarcoma cells, HT 1080, tend to detach from the culture dish coated with dimeric fibronectin shortly after reaching confluence. Lack of deposited fibronectin in the cell layer of HT 1080 might account for the detaching tendency, since normal human lung fibroblasts that do not easily detach deposited fibronectin in the cell layer. We here report that the multimeric fibronectin formed in vitro by DTT treatment at 37°C[Sakai, K., Fujii, T., and Hayashi, T.1994 J.Biochem. 115, 415-421^<1)>]significantly prevented the cell detachment. By culturing HT 1080 cells for a prolonged time of 48 h, most of the HT 1080 cells detached from the dish coated with dimeric fibronectin and became floating in the medium. On the other hand, the cell detachment from the dish was much delayed by coating dish with the multimeric fibronectin. The results suggested that the multimeric fibronectin had apotential of maintaining the tight adhesion of HT 1080 cells. The effect of the multimeric fibronectin on maintaining the cell attachment to the dish was observed when the multimeric fibronectin was added to the culture medium. The addition of the multimeric fibronectin to the renewing medium just before reaching confluence prevented the detachment of HT 1080 cells from the dish, while the addition of the dimeric fibronectin did not show the effect. The observation suggests that the multimeric fibronectin could contribute to stabilizing HT 1080 cell adhesion. The prominent effect of the multimeric fibronectin was observed only for the HT 1080 cells that appear to be deficient of multimerizing fibronectin.
