Maintenance of a Round Cell Shape and Physiological Responsiveness to Insulin of Mature Adipocytes by Culturing in Type I Collagen Gel

• Cellular behavior including viability, cell shape, and insulin responsiveness was comparedamong three culture systems of mature adipocytes obtained from rat adipose tissues : in type I collagen gel, on type I collagen-coated dish, and floating in culture medium. Rat mature adipocytes cultured in the type I collagen gel maintained a round shape with terminal differentiation state as long as 50 days or more. The adipocytes retained a responsiveness to insulin as shown by leptin secretion. The multicellular organization comprising type I collagen gel and mature adipocytes appears functionally and structurally to be an adipose tissue-like construct.
