ホルマリン固定したパラフィン包理組織からHaemophilus parasuisを検出する目的で,DNA抽出法とnested PCR法の開発を行った.nested PCR法の結果と,in situハイブリダイゼーション法により得られた結果との比較を行い検討した.キシレンによる脱パラフィン,プロテイネースKによる消化を行った後でnested PCRを行った場合に最適な結果が得られ,信頼性の高い検出法であることが示された.in situハイブリダイゼーション法ではH. Parasuisに自然感染した豚20頭で明瞭な陽性シグナルが検出された.ホルマリン固定パラフィン包理組織におけるH. Parasuisの検出に関して,nested PCR法とin situハイブリダイゼーション法での結果の一致率は100%であった.ホルマリン固定パラフィン包埋組織を用いたnested PCR法は,H. Parasuisの検出に,細菌分離検査と合わせて十分適用可能であると考えられた.
