Light-Regulated Expression of the Gene for C_4-Form Phosphoenolpyruvate Carboxylase in Maize : Destabilization of mRNA by Okadaic Acid

In Zea mays the transcripts of the phosphoenolpyruvate carboxylase (PEPC) gene for C_4 photosynthesis (Ppc1A) is known to be accumulated during greening of etiolated leaves in response to light. Effects of several kinds of inhibitors on this light-induction were investigated. The accumulation of Ppc1A mRNA was almost completely inhibited by 0.1 μM okadaic acid (OA), a protein phosphatase (PP)1 and PP2A inhibitor as revealed by northern blot analysis. In contrast, staurosporine (ST) and K252a, protein kinase inhibitors, were not effective. Diurnal change of Ppc1A mRNA level in green leaves was also collapsed by 0.1 μM OA. Accumulation of the transcripts of light harvesting chlorophyll a/b binding protein genes(Cab) during greening was also inhibited by OA, but that of the gene for a non-photosynthetic C_3-form PEPC(Ppc1B) was not. Run-on transcription analysis showed that though the transcriptional rates of Ppc1A and Cab were increased by exposure of etiolated seedlings to light, their rates were not affected by 1 μM OA. Taken together, it was proposed that a certain PP is involved in the high-level accumulation of Ppc1A mRNA under light condition through stabilization of its transcripts.
