Effects of Gabaculine on Phytochrome Synthesis during Imbibition in Embryonic Axes of Pisum sativum L.

Effects of gabaculine, a tiansaminase inhibitor, on phytochrome synthesis in the embryonic axes of Pisum sativum during imbibition at 25℃ on 0.2% agar medium were investigated. The content of phytochrome in crude homogenates (2kS) prepared from embryonic axes was determined both by a spectrophotometric assay, with highly purified phytochrome solution as the internal standard, and by an enzyme-linked immunosorbent assay (ELISA) (a sandwich method) with polyclonal and monoclonal antibodies against pea phytochrome. The content of optically detectable phytochrome in 2kS prepared from 12 h-imbibed embryonic axes was reduced in the presence of gabaculine at concentrations of 0.002 mM or higher. The maximum inhibitory effect occurred at ca. 0.1 mM. The effect of 0.5 mM gabaculine on optically detectable phytochrome became noticeable 3 h after the start of imbibition. In contrast, the time course of the increase of apophytochrome during imbibition was unaffected in the presence of gabaculine at concentrations below 1 mM. We conclude that the appearance of holophytochrome in imbibed embryonic axes results from de novo synthesis of both the protein moiety of phytochrome and its chromophore.
