Uninucleate microspores of Lilium longiflorum from different lengths of buds were explanted under various culture conditions, and their behavior was studied during sub-sequent cultures. The nutritional conditions permitting survival of microspores were relatively simple, but most of the living cells showed cytological abnormalities. A typical type of cell division could only be induced in explanted microspores at the late G_1 phase of the cell cycle. For the normal development of microspores in culture, the microspore environment with respect to moisture was an important factor. The rate of the mitotic cycle of the cultured microspores was essentially the same as that for microspores grown under greenhouse conditions.
