A transcription inhibitor in non-histone proteins of germinated pea cotyledon

A factor inhibiting in vitro transcription supported by E. coli RNA polymerase was found in isolated chromatin of germinated pea cotyledons but not in that of ungerminated ones. The factor was associated with the reconstituted chromatin and inhibited the transcription of calf thymus DNA or chromatin from ungerminated cotyledons as the template. The inhibition was not due to degradation of synthesized RNA by the factor. When the chromatin components of germinated cotyledons were dissociated in the presence of 3 M NaCl and fractionated by Bio-Gel A50, the factor appeared in the protein fractions. Further fractionation of the proteins using Bio-Rex 70 showed that the factor was contained in the non-histone protein fraction. The factor purified by Sephadex G-200 gel chromatography had an approximate molecular weight of 50,000 and was inactivated by heating at 55-60℃ for 10 min. The inhibitory effect of the purified factor was neither connected with DNase activity nor mitigated by the increasing concentrations of RNA polymerase during the assay.
