Long chain fatty acid synthesis in developing castor bean seeds IV. The synthetic system in proplastids

The proplastid fraction containing no cytosol and mitochondrion was isolated from developing castor bean endosperm by stepwise sucrose density centrifugation. This fraction possesses the capacity to synthesize LFAs from [u-^<14>C]sucrose, [u-^<14>C]-glucose, [u-^<14>C]G-1-P, [u-^<14>C]G-6-P, [2-^<14>C]pyruvate and [1-^<14>C]acetate. Little was incorporated from [1-^<14>C]pyruvate into LFAs, but much into ^<14>CO_2. Addition of cytosol to the proplastid fraction did not enhance the LFA synthesis. From these data, the whole path from sucrose to LFAs through glycolytic path and pyruvate decarboxylation seems to be located within the proplastid in developing castor bean endosperm. The difference in utilization of substrates indicates that the rate of LFA synthesis in castor bean proplastids is limited at a step between sucrose and hexose phosphate. In addition, experiments with CO_2 output and LFA synthesis from [1-^<14>C]glucose, [6-^<14>C]glucose and [u-^<14>C]G-6-P strongly suggest that the path flow branches actively through G-6-P to the pentose phosphate path and little through acetylCoA to the TCA cycle.
