Some characteristics of crude mycolytic enzymes produced by Coprinus macrorhizus f. microsporus Hongo (Z-1) and C. radians (Desm.) Fr. (Z-5) were investigated.These enzymes hardly decomposed bacterial cells, but well decomposed some sorts of yeast cells and fungal mycelia except Mucor and Rhizopus.The enzymes were also able to hydrolyse casein and laminarin, but could hardly attack paramylon, CMC-Na, pustulan, mannan and chitin. The digestion of casein was due to semi-alkaline protease with the optimum pH value of 8.5-9.0 and the digesion of laminarin resulted from β-1 : 3-glucanase with the optimum pH value of about 5.0. Therefore, these two mycolytic enzymes seem to be composed mainly of semi-alkaline protease and β-1 : 3-glucanase. This glucanase was supposed to be of an exo type.The effect of the concentration of salts in the reaction mixture on the lytic activity of the enzymes were not significant, therefore, for instance, 90% of the activity of the enzyme (Z-5) and 70% of that of the enzyme (Z-1) remained even at a concentration higher than 0.2 mole of NaCl.The activity of the enzymes was inhibited by Hg^<++> and Cu^<++> ions but not by the other heavy metal ions tested. The activity of Z-1 was inhibited by pCMB, too.As the result of partial purification of these two enzymes by a treatment with ion-exchange cellulose, the enzymes were separated into two fractions, that is, an AA fraction with strong β-1 : 3-glucanase and weak proteolytic activities and an AB fraction with a strong proteolytic activity.These two fractions could decompose heat-processed yeast cells to almost the same degree. On the other hand, the AA fraction fairly well hydrolysed lyophilized yeast cells, whereas the AB fraction scarcely did. The combination of both fractions showed a synergistic action.These results suggests that the hydrolysis of yeast cells by enzymes of Coprinus comes from semi-alkaline protease in the presence of β-1 : 3-glucanase.
