Production of Monoclonal Antibody by Hybridoma Cells Entrapped by Alginate Gel in a Medium Containing Divalent Cations and Glucose-1-Phosphate

To supply divalent cations, such as calcium or strontium ion to sustain the hardness of alginate gel, glucose-1-phosphate (G-1-P) was adopted as a phosphorus source for hybridoma cell cultures. Phosphate ions easily bind with divalent cations and precipitate them, while G-1-P does not. Hybridoma cells proliferated in a medium containing G-1-P instead of phosphate ions. Up to 10mM of strontium ions did not inhibit the growth of hybridoma cells. Hybridoma cells immobilized in alginate gel produced monoclonal antibody for 50d, while ordinary RDF medium was used alternately with the medium proposed in this article, at the same production rate as that by cells cultivated in ordinary RDF medium alone. The immobilized gel cultured in this manner was 4 times as hard and 14 times as stable as that cultivated in ordinary RDF medium.
