Trigger of Damage by Oxygen Deficiency to the Acid Production System during Submerged Acetic Fermentation with Acetobacter aceti

In order to clarify the mechanism of the phenomenon that acid productivity of Acetobacter aceti is almost completely damaged by oxygen deficiency for a short time during submerged fermentation accumulating a total acid concentration of 6% in the medium, the respiratory activity and the activities of enzymes involved in acid production of the damaged cells and the stabilities of enzymes extracted from the non-damaged cells were investigated.Respiratory activities for ethanol and acetaldehyde, and alcohol and aldehyde dehydrogenase activities, of the damaged cells decreased 20,0,20 and 50% compared to the controls, respectively.Alcohol dehydrogenase activity in the crude enzyme preparation solubilized with Triton X-100 from the membrane fraction was highly stable in culture broth containing a total acid concentration of 4.0% or less, but became quickly unstable in culture broth containing a total acid concentration exceeding 4.0%. The purified enzyme was very unstable even in culture broth of 2% total acid concentration. It was suggested from this fact that the alcohol dehydrogenase had a membrane component contributing to stability of itself. These phenomena were not observed with respect to aldehyde dehydrogenase in the membrane-solubilized preparation and the purified enzyme. Low stabilities of these enzymes in ethanol were also observed in the presence of acid of a concentration of 4% and more, compared with those in the cases of concentrations of 3% and less.
