Both tracheary element(TE) differentiation and proliferation of the mesophyll cells Zinnia elegans are suppressed in low cell density cultures with indication of cell-to-cell interaction involvement. We have investigated the effects of a sulfated pentapeptide, phytosulfokine-α(PSK-α), orginally isolated as a growth factor from asparagus suspension culture, on differentiation and proliferation of Zinnia mesophyll cells cultured at low density. When cells were incubated at an initial density of 2.5x10^4 cells/ml in TE inductive medium containing NAA(0.1mg/l) and (6-BA(1.0mg/l) in the presence of various concentrations of PSK-α, differentiation was strongly stimulated in dose-dependent fashion;more than 30% of the living cells were differentiated into TEs by the 4th day of culture in the presence of 10 nM of the factor . PSK-α also promoted proliferation of Zinnia cells in medium specified for cell division containing NAA (0.1mg/l) and 6-BA (0.1mg/l), at concentrations of 3.2nM and above. ELISA and mass spectrometry further confirmed that cultured Zinnia mesophyll cells produce nanomolar levels of PSKs under these conditions. This is the first study to demonstrate that a peptide factor riggers both cell differentiation and proliferation in plant cells.
