It has been reported that taurine exhibits a significant antihypertensive effect on essential hypertension, probably due to its natriuretic action, in which the kallikrein-kinin system may be involved. In order to clarify the effect of taurine on the renal excretion of water and sodium, sixty male SD rats, seven weeks old, were divided into three groups according to diets ; normal sodium diet (Na^+ 0.17mEq/g), high sodium diet (Na^+ 0.51mEq/g) and low sodium diet (Na^+ 0.04mEq/g). The rats belonging to each group were further divided into two groups. One was given distilled water (group C) and the other was given distilled water containing 3% taurine (group T). They were maintained for four weeks at constant temperature and humidity with exposure to light for fixed periods of time. Urinary sodium excretion, urinary volume and urinary kallikrein activity were determined by the metabolic cages at one week interval. Urinary sodium excretion and urinary volume in group T were significantly higher than in group C after two weeks. Urinary kallikrein activity in group T, which seems to reflect the production of renal kallikrein, was significantly higher than in group C after one week. These natriuretic and diuretic effects of taurine were blocked by the administration of aprotinin (50,000 KIU twice a day for four days), which is an inhibitor of kallikrein, and indomethacin (2 mg/kg for four days), which is an inhibitor of cyclooxygenase. Thus, it is concluded that taurine enhances the urinary excretion of sodium and water, which may be mediated by the increase in renal prostaglandin release caused by the activation of renal kallikrein-kinin system.
