Studies on the Infectious DNA from Bacterial Virus φX174 and from the Virus-Infected Cells

1. Heat-treated φX174 and DNA preparations obtained from φX174 by two different methods were infective to various bacterial spheroplasts including resistant strains to intact φX174 particle. 2. The subviral preparations maintained their infectivity after treatments with various proteolytic enzymes, while small amount of DNase completely destroyed the spheroplast-infectivity, indicating that the active agent was DNA itself. 3. The infectious DNA was inactivated by reacting with formaldehyde. In thermal inactivation profile, melting point was not observed. The DNA was strongly adsorbed on a methylated albumin column and eluted at a high salt concentration in the same fraction as heat-denatured E. coli DNA. These results were all consistent with the properties of single-stranded DNA molecule. 4. Infectious DNAs were also extracted from E. coli cells infected with φX174. Methylated albumin chromatography revealed that a new infectious component, different from single-stranded φX DNA, appeared in infected cells.
