The pathogenesis of Graves' disease is not known. A number of studies have shown that a great part of action of TSH on thyroid metabolism is secondary to its activation of adenylate cyclase-cyclic AMP system of thyroid glands. Attempts were made to study whether thyroidal cyclic AMP response to thyroid stimulators (TSH and PGE<SUB>1</SUB>) is different between normal human thyroids and thyroids of thyrotoxic patients. TSH was less effective in toxic thyroid slices, whereas PGE<SUB>1</SUB> was equally effective in normal and toxic thyroid slices. This difference of both kinds of tissue could not be demonstrated in normal and toxic thyroid plasma membranes. It is not known whether this less sensitivity to TSH in toxic thyroid glands is due to primary abnormality of toxic thyroid glands themselves or not.
