ON THE NEW METHODS FOR THE STAINING OF MAST CELL

The mast cells are stainable by metachromatic method and their cell granules are generally recognized as containing heparin (McMannus, 1954; Studer, 1954). Heparin is one of sulfo-mucopolysaccharides and its chemical structure is closely resemble to chondroitin sulfuric acid, though heparins obtained from different biological sources are said to be often different from each other and their chemical structures are not yet decidedly determined. The metachromatic reaction (Lison, 1953) which is often used for the histochemical demonstration of mast cell is also positive to many other negative-ionizing substances such as chondroitin sulfuric acid. This reaction is, therefore, not specific for heparin, i. e. mast cell. So the present authors, paying, attention to the Jorpes and others' report (1948) that the heparin is a sulfuric acid ester of polysaccharide, devised the following two methods for the staining of mast cell.
