Peripheral blood mononuclear cells (PBMC) from patients with autoimmune hepatitis (AIH), primary biliary cirrhosis (PBC) and healthy individuals were recostituted into sever combined immunodeficient (SCID) mice to investigate human cytokine genes expression in the mice liver. Production of human immunoglobulins (Ig) in SCID mice reconstituted with AIH-PBMC was significantly (p<0.05) increased compared to SCID mice reconstituted with PBC-PBMC and normalPBMC. Infiltrating cells around the portal area were found in all PBMC reconstituted mice, suggesting presence of the graft-versus-host (GVH) reaction. SCID mice liver reconstituted with PBC-PBMC showed severe infiltration with bile duct lesions. Using reverse transcription polymerase chain reaction (RT-PCR), interferon (IFN)-γ gene was detected in all SCID livers, while interleukin (IL)-6 and IL-1β were detected in some SCID livers. IL-6 gene correlated with human Ig levels in SCID sera, whereas increased IL-1β was found in liver with severe bile duct lesions.<BR>Our findings suggest that IFN-γ may have an important role in GVH like reaction and IL-1β may be associated with bile duct lesions in this SCID model.
