Application of Carbon Tetrachloride-Induced Liver Lesion in Mice for Screening of Liver Protective Crude Drugs

Carbon tetrachloride (CCl_44) has been orally administered to mice of nine different strains. Activities of serum glutamic-oxalacetic transaminase (SGOT) and serum glutamic-pyruvic transaminase (SGPT) have been measured 24 hr after CCl_4 administration. Dose-dependent increases of SGOT and SGPT activities have been observed in all strains. The levels of these enzyme activities in mice given the same dose of CCl_4 have varied significantly depending on the strains, indicating that the utility of mice for the screening of liver protective drugs is different among these strains. Several experiments to devise the best screening conditions have been carried out to establish mice of the strain most susceptible to CCl_4 toxicity. Since methionine and glycyrrhizin which are effective in the treatment of human hepatitis, have been found to inhibit elevation of SGOT and SGPT levels, this method is concluded to be useful as a primary screen for drugs used in human hepatitis. Using this experimental protocol, 18 crude drugs alleged to be effective against liver diseases have been examined. As a result, the buds of Artemisia capillaris, the fruits of Gardenia jasminoides, the rosettes of Artemisia capillaris, the roots of Achyranthes fauriei and the rhizomes of Curcuma longa have been found to elicit significant protective effects.
