Assay Method for Antihepatotoxic Activity Using Ionophore A23187-Induced Cytotoxicity in Primary Cultured Hepatocytes

vConditions were investigated to establish an in vitro assay method for antihepatotoxic activity using the ionophore A23187-induced cytotoxicity in primary cultured rat liver cells. Utilizing 1.5 hr preincubated rat hepatocytes, satisfactory assay procedure was achieved. Some natural products known to exert liver-protective effects in vivo were subjected to screening for antihepatotoxic activity by this in vitro assay method to reveal that glycyrrhetinic acid, glycyrrhizin and cynarin exhibited prominent antihepatotoxic activity and also significant inhibitory activity on Ca^<2+> influx into hepatocytes.
