Morphological Studies on the Pollen Grains of the Chinese Crude Drug "Huaihua"

Electron microscopic studies on the pollen grains of some plants of genus Sophora and commercially available Chinese crude drug called "Huaihua" were performed to give the following results. In order to confirm the origin of "Huaihua" in market, the pollen grains collected from the buds of Sophora japonica at similar growth stage were subjected to microscopic examination. The exterior and interior configurations of thus collected pollen grains of S. japonica were found to be very similar to the respective characteristics of "Huaihua." From the facts given above, the identity of the plant, from which "Huaihua" in market is derived, and S. japonica was established. The immatured pollen grains from "Huaihua" were found to have the same characteristics as the matured pollen grains of Sophora japonica. Thus it is now possible to identify the pollen grains of the commercially available "Huaihua." Both Sophora japonica and Pueraria spp. belong to a same family and have a same pollen type. However it is very interesting to see from the taxonomical view point that the germinal apertures of the former pollen grains were composed of three layers, namely endexine, medine and intine, on the other hand the germinal apertures of the latter were composed of two layers, namely footlayer and intine. In addition, morphological studies on the pollen grains of Sophora flavescens, S. microphylla and S. tetraptera were also performed.
