The Lignification Index (L. Index) of Bupleurum falcatum L., defined as the ratio of the area of the lignified xylem to the area of transverse section of root, was found to be higher towards the root top, lower towards the tip and about the same between 1.5-2.0 cm from the root top. When the Tsukuba, Izu and Wakayama strains of B. falcatum were cultivated in Wakayama, the plants from the seeds of Wakayama and Tsukuba strains gave a higher L. Indices than those from the seeds of Izu strain. When six strains of B. falcatum of wild origin collected in Mishima, Hiraodai, Akiyoshidai, Yufuin, Kirishima and Aso in Japan were cultivated in Nagasaki together with the above cultivated strains. With the exception of the Aso strain plants, none had obvious lignification in the root. However, higher L. Indices were shown when the seeds of these strains from the Nagasaki experimental garden were cultivated in Wakayama. These results indicate that not much lignification occur in any strain when subcultivated in warm districts, even after flowering. Further, the saikosaponin contents of this plant were found to have a negative correlation coefficient with L. Index.
