OBJECTIVE EVALUATION OF DERMAL IRRITANT BY DETERMINATION OF PROLINE HYDROXYLASE ACTIVITY IN RAT SKIN

An assay system to estimate skin irritability by chemical substances was investigated focussing on the connective tissue proliferation accompanied by skin inflammation. The proliferation rate of the connective tissue was examined by measuring the activity of proline hydroxylase, one of the enzymes for collagen metabolism, as an indicator. The dorsal neck skin of Wistar rats was coated with 2, 4-dinitrochlorbenzen and laurylbenzenesulfonate for 4 successive days, and on the 5th day the enzyme activity in the skin was measured. The proline hydroxylase in the part of the skin coated with the irritants showed clearly higher activity than that of normal skin, although it was still lower than the injured skin region prepared as a positive control. In this experiment two methods were investigated in order to extract the enzyme: i.e. one was to homogenize the whole skin, and the other to shave off the skin with the knife. The latter proved to be very efficient for enzyme extraction, and the assay of proline hydroxylase activity proved to be one of the useful methods to determine the skin irritability.
