ENZYME INDUCTION IN LIVER MICROSOMES AND ALTERATION OF BENZO(α)PYRENE MATABOLISM IN ISOLATED LIVER CELLS FROM RATS ADMINISTERED FOOD ADDITIVES

Rats were administered several food additives, and then the paramenters of the enzyme inducition in the liver microsome such as contents of P450, and the activities of aryl hydrocarbon hydroxylase(benzo(α)pyrene hydroxylase), aniline hydroxylase, aminopyrine-N-demethylase and NADPH-cytochrome c reductase, were examined. Lipid-soluble substances such as dibutylhydroxytoluene (BHT), butylhydroxyanisol (BHA), ionone and diphenyl induced variances in the P450 content and the activities of the enzyme. BHT and ionone were relatively storong inducers of the above substances. Water-soluble benzylpropionate showed no parameter variance. Though the pattern of induction by BHT somewhat resembled that of phenobarbital (PB), that of ionone could not be classified. CO-complex difference spectra of the microsomes from the rats administered food additives were the same as those with PB. Some parameters showed that food additives induced individual variances in the effects. Further, the metabolism of carcinogen benzo(α)pyrene was exmained in the isolated liver cells. The hydroxylation of benzo(α)pyrene in the cell was detected more markedly by histofluorometry in the cytoplasm than in the nucleus. The viable liver cells from the rats administered BHT and ionone showed 2.4- and 2.9-fold greater benzo(α)pyrene-hydroxylation activity, respectively, than the control. Therefore, some food additives induce microsomal drug metabolizing enzymes, and hence they might affect the metabolism of the carcinogen.
